Sermorelin
About
About Sermorelin
A synthetic analog of growth hormone-releasing hormone (GHRH), this 29-amino acid peptide stimulates the pituitary gland to produce and secrete endogenous growth hormone through binding to GHRH receptors. It functions by mimicking the N-terminal bioactive portion of native GHRH, promoting pulsatile GH release while preserving the body's natural feedback mechanisms. Primary applications include diagnostic testing of pituitary function and therapeutic use in growth hormone deficiency.
Science
Mechanism of Action
Sermorelin binds to the GHRH receptor (GHRHR) on pituitary somatotrophs, stimulating adenylyl cyclase and increasing intracellular cAMP. This triggers calcium influx and GH vesicle exocytosis. The downstream effects — elevated serum GH followed by hepatic IGF-1 production — produce anabolic, lipolytic, and regenerative effects in peripheral tissues. Sermorelin's pulsatile mechanism preserves natural GH release patterns better than exogenous HGH administration.
Dosing
Typical Protocol
200–500 mcg subcutaneously before sleep daily. Monitor IGF-1 at baseline and after 8–12 weeks. Protocols typically run for 3–6 month cycles. Can be combined with Ipamorelin for enhanced GH pulse amplitude. Best administered on an empty stomach to avoid competition with glucose-stimulated insulin.

Regulatory
Legal Status in 2026
This compound is legal to prescribe and dispense through a licensed 503A or 503B compounding pharmacy with a valid prescription. It is not FDA-approved as a finished drug product, but compounding is explicitly permitted under federal law. Quality and sterility standards vary by pharmacy — look for PCAB-accredited compounders.
Evidence
Evidence Tier
Evidence from randomized controlled trials (RCTs) or large observational studies in humans. This is the gold standard — effects have been measured in controlled conditions and results are peer-reviewed and reproducible.
